چکیده

We present a new super-resolution technique, Re-scan Confocal Microscopy (RCM), based on standard confocal microscopy extended with an optical (re-scanning) unit that projects the image directly on a CCD-camera. This new microscope has improved lateral resolution and strongly improved sensitivity while maintaining the sectioning capability of a standard confocal microscope. This simple technology is typically useful for biological applications where the combination high-resolution and high-sensitivity is required.

Optimization of pupil design for point-scanning and line-scanning confocal microscopy

Both point-scanning and line-scanning confocal microscopes provide resolution and optical sectioning to observe nuclear and cellular detail in human tissues, and are being translated for clinical applications. While traditional point-scanning is truly confocal and offers the best possible optical sectioning and resolution, line-scanning is partially confocal but may offer a relatively simpler a...

High-speed multifocal array scanning using refractive window tilting.

Confocal microscopy has several advantages over wide-field microscopy, such as out-of-focus light suppression, 3D sectioning, and compatibility with specialized detectors. While wide-field microscopy is a faster approach, multiplexed confocal schemes can be used to make confocal microscopy more suitable for high-throughput applications, such as high-content screening (HCS) commonly used in drug...

Optical photon reassignment with increased axial resolution by structured illumination.

Fluorescent microscopy methods linked to the reassignment principle as image scanning microscopy (ISM), re-scan confocal (RSC), optical photon reassignment (OPRA) and instant structured illumination microscopy (iSIM) have the potential to replace confocal microscopy as the standard microscopy technique. Photon reassignment methods are known to link the most important properties in biological im...

Scanning and Image Reconstruction Techniques in Confocal Laser Scanning Microscopy

Taking advantage of the laser scanning methodology, confocal microscopy has been widely applied in cutting-edge biological research for its three-dimensional, high-resolution imaging capability (Pawley 1995). The principles of confocal were put forward by Marvin Minsky in 1955 (Minsky 1988), yet the experimental demonstration was not accomplished until 20 years later by Cremer brother (Cremer, ...

Optical axial scanning in confocal microscopy using an electrically tunable lens.

This paper presents the use and characterization of an electrically focus tunable lens to perform axial scanning in a confocal microscope. Lateral and axial resolution are characterized over a >250 µm axial scan range. Confocal microscopy using optical axial scanning is demonstrated in epithelial tissue and compared to traditional stage scanning. By enabling rapid axial scanning, minimizing mot...
